This variant lies in the CLCN1 gene (transcript NM_000083.3) at coding-DNA position 1699, where A is replaced by C; at the protein level this means replaces asparagine at residue 567 with histidine — a missense variant. Submitter rationale: This sequence change replaces asparagine, which is neutral and polar, with histidine, which is basic and polar, at codon 567 of the CLCN1 protein (p.Asn567His). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with autosomal recessive myotonia congenita (PMID: 23113340). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant.
