Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_001283009.2(RTEL1):c.1030C>G (p.Pro344Ala), citing Ambry Variant Classification Scheme 2023: The p.P344A variant (also known as c.1030C>G), located in coding exon 11 of the RTEL1 gene, results from a C to G substitution at nucleotide position 1030. The proline at codon 344 is replaced by alanine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
