Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_000138.5(FBN1):c.1262C>T (p.Pro421Leu), citing Ambry Variant Classification Scheme 2023: The p.P421L variant (also known as c.1262C>T), located in coding exon 10 of the FBN1 gene, results from a C to T substitution at nucleotide position 1262. The proline at codon 421 is replaced by leucine, an amino acid with similar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
