This variant lies in the PARN gene (transcript NM_002582.4) at coding-DNA position 1667, where A is replaced by G; at the protein level this means replaces asparagine at residue 556 with serine — a missense variant. Submitter rationale: This sequence change replaces asparagine, which is neutral and polar, with serine, which is neutral and polar, at codon 556 of the PARN protein (p.Asn556Ser). This variant is present in population databases (rs762930640, gnomAD 0.006%). This variant has not been reported in the literature in individuals affected with PARN-related conditions.
