Uncertain significance for Pheochromocytoma/paraganglioma syndrome 5; Mitochondrial complex II deficiency, nuclear type 1 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_004168.4(SDHA):c.539T>C (p.Phe180Ser), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the SDHA gene (transcript NM_004168.4) at coding-DNA position 539, where T is replaced by C; at the protein level this means replaces phenylalanine at residue 180 with serine — a missense variant. Submitter rationale: This sequence change replaces phenylalanine, which is neutral and non-polar, with serine, which is neutral and polar, at codon 180 of the SDHA protein (p.Phe180Ser). This variant is not present in population databases (gnomAD no frequency). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive. This variant has not been reported in the literature in individuals affected with SDHA-related conditions.
