Uncertain significance for Progressive myoclonic epilepsy type 8 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_021267.5(CERS1):c.940G>T (p.Val314Leu), citing Invitae Variant Classification Sherloc (09022015): An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be tolerated. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. This variant has not been reported in the literature in individuals affected with CERS1-related conditions. This sequence change replaces valine, which is neutral and non-polar, with leucine, which is neutral and non-polar, at codon 314 of the CERS1 protein (p.Val314Leu). This variant is present in population databases (no rsID available, gnomAD 0.002%).
