NM_017849.4(TMEM127):c.278T>G (p.Leu93Arg) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.L93R variant (also known as c.278T>G), located in coding exon 2 of the TMEM127 gene, results from a T to G substitution at nucleotide position 278. The leucine at codon 93 is replaced by arginine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
