Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_178138.6(LHX3):c.203G>A (p.Arg68His), citing Invitae Variant Classification Sherloc (09022015): This variant has not been reported in the literature in individuals affected with LHX3-related conditions. This variant is present in population databases (rs377183234, gnomAD 0.002%). This sequence change replaces arginine, which is basic and polar, with histidine, which is basic and polar, at codon 73 of the LHX3 protein (p.Arg73His). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be disruptive. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
