Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_004541.4(NDUFA1):c.176G>A (p.Arg59His), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces arginine, which is basic and polar, with histidine, which is basic and polar, at codon 59 of the NDUFA1 protein (p.Arg59His). This variant is present in population databases (rs746256856, gnomAD 0.002%), including at least one homozygous and/or hemizygous individual. This variant has not been reported in the literature in individuals affected with NDUFA1-related conditions. ClinVar contains an entry for this variant (Variation ID: 2912795). An algorithm developed to predict the effect of missense changes on protein structure and function outputs the following: PolyPhen-2: "Benign". The histidine amino acid residue is found in multiple mammalian species, which suggests that this missense change does not adversely affect protein function. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
