Uncertain significance for Primary ciliary dyskinesia — the classification assigned by Ambry Genetics to NM_001277115.2(DNAH11):c.9250C>G (p.Leu3084Val), citing Ambry Variant Classification Scheme 2023: The p.L3084V variant (also known as c.9250C>G), located in coding exon 56 of the DNAH11 gene, results from a C to G substitution at nucleotide position 9250. The leucine at codon 3084 is replaced by valine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this alteration remains unclear.
