Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_003998.4(NFKB1):c.1214A>G (p.Tyr405Cys), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the NFKB1 gene (transcript NM_003998.4) at coding-DNA position 1214, where A is replaced by G; at the protein level this means replaces tyrosine at residue 405 with cysteine — a missense variant. Submitter rationale: This sequence change replaces tyrosine, which is neutral and polar, with cysteine, which is neutral and slightly polar, at codon 405 of the NFKB1 protein (p.Tyr405Cys). This variant is present in population databases (rs573164232, gnomAD 0.006%). This variant has not been reported in the literature in individuals affected with NFKB1-related conditions. An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be disruptive. Experimental studies have shown that this missense change does not substantially affect NFKB1 function (PMID: 36105815). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
