This variant lies in the APC gene (transcript NM_000038.6) at 6 bases into the intron immediately after coding-DNA position 135 through 7 bases into the intron immediately after coding-DNA position 135, deleting this region. Submitter rationale: This variant deletes two nucleotides at position +6 and +7 in intron 2 of the APC gene. Splice site prediction tools suggest that this variant may not impact RNA splicing. To our knowledge, RNA studies have not been reported for this variant. This variant has not been reported in individuals affected with APC-related disorders in the literature. This variant has not been identified in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.
