This variant lies in the MYO7A gene (transcript NM_000260.4) at coding-DNA position 1954, where T is replaced by C; at the protein level this means replaces cysteine at residue 652 with arginine — a missense variant. Submitter rationale: Variant summary: MYO7A c.1954T>C (p.Cys652Arg) results in a non-conservative amino acid change located in the Myosin. Large ATPases domain (IPR001609) of the encoded protein sequence. Five of five in-silico tools predict a damaging effect of the variant on protein function. The variant was absent in 227028 control chromosomes. c.1954T>C has been reported in trans with a pathogenic frameshift in the literature in multiple siblings affected with clinical features of Usher Syndrome (example, Wang_2017). These data indicate that the variant may be associated with disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
