This variant lies in the ABCA12 gene (transcript NM_173076.3) at coding-DNA position 5884, where G is replaced by A; at the protein level this means replaces glycine at residue 1962 with serine — a missense variant. Submitter rationale: This sequence change replaces glycine, which is neutral and non-polar, with serine, which is neutral and polar, at codon 1962 of the ABCA12 protein (p.Gly1962Ser). This variant also falls at the last nucleotide of exon 39, which is part of the consensus splice site for this exon. This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with ichthyosis (PMID: 29298786, 32978145). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be tolerated.
