Uncertain Significance for Ehlers-Danlos syndrome, type 4 — the classification assigned by All of Us Research Program, National Institutes of Health to NM_000090.4(COL3A1):c.582+6dup, citing ACMG Guidelines, 2015. This variant lies in the COL3A1 gene (transcript NM_000090.4) at 6 bases into the intron immediately after coding-DNA position 582, duplicating one base. Submitter rationale: This variant causes duplication of a nucleotide in intron 6 of the COL3A1 gene. To our knowledge, functional studies have not been reported for this variant. This variant has not been reported in individuals affected with COL3A1-related disorders in the literature. This variant has not been identified in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.

This study involves interpretation of variants in research participants for the purpose of population health screening. Participant phenotype was not available at the time of variant classification. Additional details can be found in publication PMID: 35346344, PMCID: PMC8962531
