This variant lies in the ABCC2 gene (transcript NM_000392.5) at coding-DNA position 1321, where C is replaced by A; at the protein level this means replaces leucine at residue 441 with methionine — a missense variant. Submitter rationale: Variant summary: ABCC2 c.1321C>A (p.Leu441Met) results in a conservative amino acid change located in the ABC transporter type 1, transmembrane domain (IPR011527) of the encoded protein sequence. Four of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 4e-06 in 251240 control chromosomes (gnomAD). c.1321C>A has been reported in the literature in individuals affected with Dubin-Johnson Syndrome (Lee_2006, Chen_2019).
