Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000255.4(MMUT):c.2193dup (p.Ala732fs), citing Invitae Variant Classification Sherloc (09022015): This sequence change creates a premature translational stop signal (p.Ala732Cysfs*6) in the MUT gene. While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 19 amino acid(s) of the MUT protein. This variant is present in population databases (rs747331003, gnomAD 0.003%). This premature translational stop signal has been observed in individual(s) with methylmalonic acidemia (PMID: 10923046, 16281286, 16435223, 27167370). This variant is also known as 2270del4/ins5 and c.2194-2197del4ins5 (p.Ala732fsX737). Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site. For these reasons, this variant has been classified as Pathogenic.
