The variant allele was found at a frequency of 0.00029 in 251370 control chromosomes, predominantly at a frequency of 0.0039 within the East Asian subpopulation in the gnomAD database. The observed variant frequency within East Asian control individuals in the gnomAD database exceeds the estimated maximal expected allele frequency for disease-causing variants in BSND. c.16A>G has been observed with a variant in another gene in an individual affected with hearing loss (Liu_2022). These report(s) do not provide unequivocal conclusions about association of the variant with Bartter Syndrome, Type 4a. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
