This variant disrupts a region of the SCN1A protein in which other variant(s) (p.Arg1912*) have been determined to be pathogenic (PMID: 14738421, 20522430, 23195492; Invitae). This suggests that this is a clinically significant region of the protein, and that variants that disrupt it are likely to be disease-causing. This premature translational stop signal has been observed in individual(s) with SCN1A-related conditions (PMID: 31487502). In at least one individual the variant was observed to be de novo. This variant is not present in population databases (gnomAD no frequency). This sequence change creates a premature translational stop signal (p.Lys1827Asnfs*31) in the SCN1A gene. While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 183 amino acid(s) of the SCN1A protein.
