Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_002282.3(KRT83):c.173G>A (p.Gly58Asp), citing Invitae Variant Classification Sherloc (09022015): In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. This variant has not been reported in the literature in individuals affected with KRT83-related conditions. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is not expected to disrupt KRT83 protein function. This variant is present in population databases (rs776629118, gnomAD 0.003%). This sequence change replaces glycine, which is neutral and non-polar, with aspartic acid, which is acidic and polar, at codon 58 of the KRT83 protein (p.Gly58Asp).
