This variant lies in the SYNE2 gene (transcript NM_182914.3) at coding-DNA position 13020 through 9 bases into the intron immediately after coding-DNA position 13023, deleting this region. Submitter rationale: The frequency data for this variant in the population databases is considered unreliable, as metrics indicate poor data quality at this position in the gnomAD database. This variant results in the deletion of part of exon 67 (c.13020_13023+9del) of the SYNE2 gene. It is expected to disrupt RNA splicing. Variants that disrupt the donor or acceptor splice site typically lead to a loss of protein function (PMID: 16199547), however the current clinical and genetic evidence is not sufficient to establish whether loss-of-function variants in SYNE2 cause disease.
