Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000372.5(TYR):c.1322C>A (p.Ser441Ter), citing Invitae Variant Classification Sherloc (09022015): This variant is present in population databases (rs544365966, gnomAD 0.0009%). This sequence change creates a premature translational stop signal (p.Ser441*) in the TYR gene. While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 89 amino acid(s) of the TYR protein. This variant has not been reported in the literature in individuals affected with TYR-related conditions. For these reasons, this variant has been classified as Pathogenic. This variant disrupts a region of the TYR protein in which other variant(s) (p.Ala490Cysfs*20) have been determined to be pathogenic (PMID: 1642278, 1711223, 13680365, 18463683). This suggests that this is a clinically significant region of the protein, and that variants that disrupt it are likely to be disease-causing.
