Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_002645.4(PIK3C2A):c.2723G>T (p.Cys908Phe), citing Invitae Variant Classification Sherloc (09022015): This variant has not been reported in the literature in individuals affected with PIK3C2A-related conditions. This variant is present in population databases (rs112584158, gnomAD 0.01%). This sequence change replaces cysteine, which is neutral and slightly polar, with phenylalanine, which is neutral and non-polar, at codon 908 of the PIK3C2A protein (p.Cys908Phe). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be disruptive. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site.
