NM_181486.4(TBX5):c.1010C>A (p.Pro337His) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.P337H variant (also known as c.1010C>A), located in coding exon 8 of the TBX5 gene, results from a C to A substitution at nucleotide position 1010. The proline at codon 337 is replaced by histidine, an amino acid with similar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
