This variant lies in the SMAD6 gene (transcript NM_005585.5) at coding-DNA position 998, where G is replaced by A; at the protein level this means replaces serine at residue 333 with asparagine — a missense variant. Submitter rationale: Variant summary: SMAD6 c.998G>A (p.Ser333Asn) results in a conservative amino acid change located in the SMAD, Dwarfin-type domain (IPR001132) of the encoded protein sequence. Three of five in-silico tools predict a benign effect of the variant on protein function. The variant allele was found at a frequency of 1.3e-05 in 229762 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.998G>A has been reported in the literature at least once in a cohort of individuals affected with heritable thoracic aortic disease (Musfee_2020). This report does not provide unequivocal conclusions about association of the variant with Aortic Valve Disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
