Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_004387.4(NKX2-5):c.701C>G (p.Ser234Trp), citing Ambry Variant Classification Scheme 2023: The p.S234W variant (also known as c.701C>G), located in coding exon 2 of the NKX2-5 gene, results from a C to G substitution at nucleotide position 701. The serine at codon 234 is replaced by tryptophan, an amino acid with highly dissimilar properties. This amino acid position is not well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
