Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_024598.4(USB1):c.205G>A (p.Gly69Arg), citing Invitae Variant Classification Sherloc (09022015): Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt USB1 protein function. This variant has not been reported in the literature in individuals affected with USB1-related conditions. This variant is present in population databases (rs762121009, gnomAD 0.003%). This sequence change replaces glycine, which is neutral and non-polar, with arginine, which is basic and polar, at codon 69 of the USB1 protein (p.Gly69Arg). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
