Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001130009.3(GEN1):c.1201C>T (p.Arg401Ter), citing Invitae Variant Classification Sherloc (09022015): This sequence change creates a premature translational stop signal (p.Arg401*) in the GEN1 gene. It is expected to result in an absent or disrupted protein product. However, the current clinical and genetic evidence is not sufficient to establish whether loss-of-function variants in GEN1 cause disease. This variant is present in population databases (rs747865506, gnomAD 0.05%). This premature translational stop signal has been observed in individual(s) with non-small cell lung cancer (PMID: 36113475). Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
