Pathogenic for Short-rib thoracic dysplasia 11 with or without polydactyly — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_052844.4(DYNC2I2):c.256del (p.Gln86fs), citing Invitae Variant Classification Sherloc (09022015): For these reasons, this variant has been classified as Pathogenic. This variant has not been reported in the literature in individuals affected with WDR34-related conditions. This variant is present in population databases (rs770506724, gnomAD 0.0009%). This sequence change creates a premature translational stop signal (p.Gln86Argfs*38) in the WDR34 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in WDR34 are known to be pathogenic (PMID: 24183449, 24183451, 28379358).
