Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_001148.6(ANK2):c.6595G>C (p.Asp2199His), citing Ambry Variant Classification Scheme 2023: The p.D2199H variant (also known as c.6595G>C), located in coding exon 38 of the ANK2 gene, results from a G to C substitution at nucleotide position 6595. The aspartic acid at codon 2199 is replaced by histidine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this alteration remains unclear.
