Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_001999.4(FBN2):c.7837G>A (p.Glu2613Lys), citing Ambry Variant Classification Scheme 2023: The p.E2613K variant (also known as c.7837G>A), located in coding exon 61 of the FBN2 gene, results from a G to A substitution at nucleotide position 7837. The glutamic acid at codon 2613 is replaced by lysine, an amino acid with similar properties. This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
