Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000548.5(TSC2):c.1369T>C (p.Ser457Pro), citing Ambry Variant Classification Scheme 2023: The p.S457P variant (also known as c.1369T>C), located in coding exon 13 of the TSC2 gene, results from a T to C substitution at nucleotide position 1369. The serine at codon 457 is replaced by proline, an amino acid with similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
