Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_001079802.2(FKTN):c.374G>T (p.Gly125Val), citing Ambry Variant Classification Scheme 2023: The p.G125V variant (also known as c.374G>T), located in coding exon 4 of the FKTN gene, results from a G to T substitution at nucleotide position 374. The glycine at codon 125 is replaced by valine, an amino acid with dissimilar properties. This alteration has been reported in a sudden unexplained death cohort, an ocular disorders cohort, and a dilated cardiomyopathy (DCM) cohort (Lahrouchi N et al. Eur J Hum Genet, 2020 Jan;28:17-22; Mazzarotto F et al. Circulation, 2020 Feb;141:387-398; Deml B et al. Eur J Hum Genet, 2016 Apr;24:535-41). This amino acid position is not well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
