NM_001042492.3(NF1):c.6430G>C (p.Glu2144Gln) was classified as Uncertain significance for Cardiovascular phenotype; Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.E2123Q variant (also known as c.6367G>C), located in coding exon 42 of the NF1 gene, results from a G to C substitution at nucleotide position 6367. The glutamic acid at codon 2123 is replaced by glutamine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
