Submitter rationale: The c.8755-3C>A intronic variant results from a C to A substitution 3 nucleotides upstream from coding exon 21 in the BRCA2 gene. Two saturation genome editing-based studies, including a haploid cell-survival assay and a humanized mouse embryonic stem cell line assay of drug response and survival, demonstrate that this nucleotide substitution is functional (Huang H et al. Nature. 2025 Feb;638(8050):528-537; Sahu S et al. Nature. 2025 Feb;638(8050):538-545). This nucleotide position is not well conserved in available vertebrate species. In silico splice site analysis predicts that this alteration may result in the creation or strengthening of a novel splice acceptor site. RNA studies have demonstrated that this variant results in abnormal splicing in the set of samples tested (Ambry internal data). Based on the available evidence, the clinical significance of this variant remains unclear.
