NM_001384474.1(LOXHD1):c.197C>G (p.Thr66Arg) was classified as Uncertain significance by GeneDx, citing GeneDx Variant Classification (06012015). This variant lies in the LOXHD1 gene (transcript NM_001384474.1) at coding-DNA position 197, where C is replaced by G; at the protein level this means replaces threonine at residue 66 with arginine — a missense variant. Submitter rationale: The T66R variant in the LOXHD1 gene has not been reported previously as a pathogenic variant, nor as a benign variant, to our knowledge. The T66R variant is observed in 1/2176 (0.05%) alleles from individuals of African background, in large population cohorts (Lek et al., 2016; 1000 Genomes Consortium et al., 2015; Exome Variant Server). The T66R variant is a semi-conservative amino acid substitution, which may impact secondary protein structure as these residues differ in some properties. This substitution occurs at a position where amino acids with similar properties to Threonine are tolerated across species. In silico analysis predicts this variant is probably damaging to the protein structure/function. We interpret T66R as a variant of uncertain significance.
