This variant lies in the GDAP1 gene (transcript NM_018972.4) at coding-DNA position 449, where C is replaced by T; at the protein level this means replaces serine at residue 150 with phenylalanine — a missense variant. Submitter rationale: This sequence change replaces serine, which is neutral and polar, with phenylalanine, which is neutral and non-polar, at codon 150 of the GDAP1 protein (p.Ser150Phe). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with clinical features of autosomal recessive Charcot-Marie-Tooth disease (Invitae).
