Pathogenic for Developmental and epileptic encephalopathy 94 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001271.4(CHD2):c.2134A>G (p.Lys712Glu), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the CHD2 gene (transcript NM_001271.4) at coding-DNA position 2134, where A is replaced by G; at the protein level this means replaces lysine at residue 712 with glutamic acid — a missense variant. Submitter rationale: This sequence change replaces lysine, which is basic and polar, with glutamic acid, which is acidic and polar, at codon 712 of the CHD2 protein (p.Lys712Glu). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with clinical features of CHD2-related conditions (Invitae). In at least one individual the variant was observed to be de novo. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt CHD2 protein function. For these reasons, this variant has been classified as Pathogenic.
