NM_004100.5(EYA4):c.110G>A (p.Ser37Asn) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.S37N variant (also known as c.110G>A), located in coding exon 3 of the EYA4 gene, results from a G to A substitution at nucleotide position 110. The serine at codon 37 is replaced by asparagine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
