Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001069.3(TUBB2A):c.22C>T (p.Gln8Ter), citing Invitae Variant Classification Sherloc (09022015): This sequence change creates a premature translational stop signal (p.Gln8*) in the TUBB2A gene. It is expected to result in an absent or disrupted protein product. However, the current clinical and genetic evidence is not sufficient to establish whether loss-of-function variants in TUBB2A cause disease. This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with TUBB2A-related conditions. This premature translational stop signal has been observed in at least one individual who was not affected with TUBB2A-related conditions (Invitae). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Experimental studies and prediction algorithms are not available or were not evaluated, and the functional significance of this variant is currently unknown.
