Pathogenic for Hyperkalemic periodic paralysis — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000334.4(SCN4A):c.1331C>A (p.Ala444Asp), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces alanine, which is neutral and non-polar, with aspartic acid, which is acidic and polar, at codon 444 of the SCN4A protein (p.Ala444Asp). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with paramyotonia congenita (PMID: 21220685). In at least one individual the variant was observed to be de novo. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt SCN4A protein function. For these reasons, this variant has been classified as Pathogenic.
