Pathogenic for Eichsfeld type congenital muscular dystrophy — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_206926.2(SELENON):c.788_879dup (p.Asp294fs), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the SELENON gene (transcript NM_206926.2) at coding-DNA position 788 through coding-DNA position 879, duplicating 92 bases; at the protein level this means shifts the reading frame starting at aspartic acid residue 294, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: For these reasons, this variant has been classified as Pathogenic. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site. This variant has not been reported in the literature in individuals affected with SELENON-related conditions. This variant is not present in population databases (gnomAD no frequency). This sequence change creates a premature translational stop signal (p.Asp328Serfs*43) in the SELENON gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in SELENON are known to be pathogenic (PMID: 21131290, 21670436).
