Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_005199.5(CHRNG):c.1123C>T (p.Gln375Ter), citing Invitae Variant Classification Sherloc (09022015): For these reasons, this variant has been classified as Pathogenic. This variant has not been reported in the literature in individuals affected with CHRNG-related conditions. This variant is present in population databases (rs771796518, gnomAD 0.01%). This sequence change creates a premature translational stop signal (p.Gln375*) in the CHRNG gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in CHRNG are known to be pathogenic (PMID: 16826520).
