Pathogenic for Hypertrophic cardiomyopathy — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000256.3(MYBPC3):c.165C>G (p.Tyr55Ter), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the MYBPC3 gene (transcript NM_000256.3) at coding-DNA position 165, where C is replaced by G; at the protein level this means converts the codon for tyrosine at residue 55 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: This variant is not present in population databases (gnomAD no frequency). For these reasons, this variant has been classified as Pathogenic. This variant has not been reported in the literature in individuals affected with MYBPC3-related conditions. This sequence change creates a premature translational stop signal (p.Tyr55*) in the MYBPC3 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in MYBPC3 are known to be pathogenic (PMID: 19574547).
