NM_020297.4(ABCC9):c.1266T>G (p.Asn422Lys) was classified as Pathogenic for Dilated cardiomyopathy 1O by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces asparagine, which is neutral and polar, with lysine, which is basic and polar, at codon 422 of the ABCC9 protein (p.Asn422Lys). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with clinical features of Cantu syndrome (Invitae). In at least one individual the variant was observed to be de novo. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt ABCC9 protein function. For these reasons, this variant has been classified as Pathogenic.
