This variant lies in the LMNA gene (transcript NM_170707.4) at coding-DNA position 1046, where G is replaced by A; at the protein level this means replaces arginine at residue 349 with glutamine — a missense variant. Submitter rationale: This missense variant replaces arginine with glutamine at codon 349 of the LMNA protein. Computational prediction tools indicate that this variant has a deleterious impact on protein structure and function. To our knowledge, functional studies have not been reported for this variant. This variant has been reported in at least one individual affected with dilated cardiomyopathy (PMID: 25163546, 27532257). This variant has been identified in 1/251274 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.
