Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_006231.4(POLE):c.377G>A (p.Gly126Asp), citing Ambry Variant Classification Scheme 2023: The p.G126D variant (also known as c.377G>A), located in coding exon 5 of the POLE gene, results from a G to A substitution at nucleotide position 377. The glycine at codon 126 is replaced by aspartic acid, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
