Uncertain significance — the classification assigned by GeneDx to NM_032382.5(COG8):c.1079G>A (p.Gly360Glu), citing GeneDx Variant Classification (06012015): The G360E variant in the COG8 gene has not been published as a pathogenic variant, nor as a benign variant, to our knowledge. The G360E variant was not observed with any significant frequency in approximately 6500 individuals of European and African American ancestry in the NHLBI Exome Sequencing Project. The G360E variant is a non-conservative amino acid substitution, which occurs at a position that is conserved across species. In silico analysis predicts this variant is probably damaging to the protein structure/function. Based on review of the data in the context of the 2015 ACMG standards and guidelines for the interpretation of sequence variants (Richards et al., 2015), we now interpret G360E as a variant of uncertain significance.
