NM_001042492.3(NF1):c.5269-3C>G was classified as Likely pathogenic for Cardiovascular phenotype; Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The c.5206-3C>G intronic variant results from a C to G substitution 3 nucleotides upstream from coding exon 37 in the NF1 gene. This variant was reported in individual(s) with features consistent with Neurofibromatosis type1 (Ambry internal data). This nucleotide position is not well conserved in available vertebrate species. In silico splice site analysis predicts that this alteration may weaken the native splice acceptor site. RNA studies have demonstrated that this alteration results in abnormal splicing in the set of samples tested (Ambry internal data). This variant is considered to be rare based on population cohorts in the Genome Aggregation Database (gnomAD). Based on the majority of available evidence to date, this variant is likely to be pathogenic.
