Pathogenic for Neuromuscular disease caused by qualitative or quantitative defects of dystrophin — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_004006.3(DMD):c.1594C>T (p.Gln532Ter), citing LabCorp Variant Classification Summary - May 2015. This variant lies in the DMD gene (transcript NM_004006.3) at coding-DNA position 1594, where C is replaced by T; at the protein level this means converts the codon for glutamine at residue 532 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: Variant summary: DMD c.1594C>T (p.Gln532X) results in a premature termination codon, predicted to cause absence of the protein due to nonsense mediated decay, a commonly known mechanism for disease. The variant was absent in 182717 control chromosomes (gnomAD). c.1594C>T has been reported in the literature in individuals affected with Dystrophinopathies such as Duchenne muscular dystrophy (e.g. Xu_2018). These data indicate that the variant is very likely to be associated with disease. The following publication has been ascertained in the context of this evaluation (PMID: 29604111). Two submitters have provided clinical-significance assessments for this variant to ClinVar after 2014 and both classified the variant as pathogenic. Based on the evidence outlined above, the variant was classified as pathogenic.
